Her performance in this role saw her land her first leading role in the 2004 drama film, Shreds and Dreams.
In 2005, Winnie Ntshaba began starring as Khethiwe Buthelezi in the SABC1’s soapie, Generations. In 2015 she was fired along with other she and 15 actors when they staged a strike against the showrunners in demand of an extension of their contracts and royalties from episodes which had been rebroadcast. Following her departure from Generations, Winnie Ntshaba took a three-year break from television, returning in 2018 with a leading role of Mam’Ngadi in Mzansi Magic’s drama, The Herd.
Winnie Ntshaba has also appeared in Isithembiso as Mam’Lihle and in House of Zwide as Funani Zwide’s wife.
