Masina joined ANC Youth politics at 14 years of age, he served in various positions from the street bottom before raising to regional and provincial leadership of the ANC Youth League.

- ANC Enkululekweni branch secretary and is currently a member of the ANC Ekurhuleni Regional Executive Committee.

Mzwandile Masina resignation
On 31 May 2020 Masina posted a controversial tweet, this lead to high speculation of his resignation. He threatened to resign from the mayoral post but later retracted his resignation threats.
The tweet comments were posted in solidarity with EFF Leader Julius Malema’s message that
“ Let White Economy Collapse”.
Masina tweeted his response in solidarity with brother and friend Julius Malema.
“I fully agree with my friend on this one @Julius_S_Malema. Until we nationalise all commanding heights of the economy- we need to restart this economy and make sure it favors the majority. Things can’t be sane again”
